<p>Stimulated by the 2012 Nobel Prize in Physiology or Medicine awarded for Shinya Yamanaka and Sir John Gurdon, there is an increasing interest in the iPS cells and reprogramming technologies in medical science. While iPS cells are expected to open new era providing enormous opportunities in the biomedical sciences in terms of cell therapies for regenerative medicine, safety-related concerns for iPS cell-based cell therapy should be resolved prior to the clinical application of iPS cells. In this symposium, the pre-clinical investigations of cell therapy for SCI using neural stem/progenitor cells derived from iPS cells, and their safety issues in vivo are outlined.</p>

Generation of human melanocytes from induced pluripotent stem cells

<p>The discovery of human induced pluripotent stem cells (iPSCs) has provided a model system for studying early events during human development. Developmentally melanocytes originate from migratory neural crest cells that emerge from the neural plate during embryogenesis after a complex process of differentiation, proliferation, and migration out of the neural tube along defined pathways. In the adult, human melanocytes are located in the basal layer of the epidermis, hair follicles, uvea, inner ear, and meninges. In the epidermis, melanocytes produce melanin pigment that gives color to the skin as well as providing protection from ultraviolet light damage. In addition, melanocytes transfer melanin pigment to hair matrix keratinocytes during each hair cycle to maintain hair pigmentation. Characterization of mouse melanocyte stem cells (MELSCs) is more complete than for humans. MELSCs are located in the bulge region of hair follicles, where hair follicle stem cells (HFSCs) also reside. Recently, it has been demonstrated that HFSCs provide a functional nice for MELSCs. According to current cancer stem cell theory, melanomas are considered to evolve from MELSCs, although the exact mechanism remains to be elucidated fully. In humans, importantly, the lack of more specific markers of MELSCs, current understanding of the molecular regulations of melanocyte development remains incomplete. Recently, the generation of melanocytes from iPSCs has lead to some clarification of human melanocyte development in vitro. Utilization of iPSC-derived melanocytes may prove invaluable in further study of human melanocytic development and novel therapies for patients suffering with pigmentation disorders and melanoma. © 2013 Springer Science+Business Media New York.</p>

<p>Somatic mutation of RUNX1 is implicated in various hematological malignancies, including myelodysplastic syndrome and acute myeloid leukemia (AML), and previous studies using mouse models disclosed its critical roles in hematopoiesis. However, the role of RUNX1 in human hematopoiesis has never been tested in experimental settings. Familial platelet disorder (FPD)/AML is an autosomal dominant disorder caused by germline mutation of RUNX1, marked by thrombocytopenia and propensity to acute leukemia. To investigate the physiological function of RUNX1 in human hematopoiesis and pathophysiology of FPD/AML, we derived induced pluripotent stem cells (iPSCs) from three distinct FPD/AML pedigrees (FPD-iPSCs) and examined their defects in hematopoietic differentiation. By in vitro differentiation assays, FPD-iPSCs were clearly defective in the emergence of hematopoietic progenitors and differentiation of megakaryocytes, and overexpression of wild-type (WT)-RUNX1 reversed most of these phenotypes. We further demonstrated that overexpression of mutant-RUNX1 in WT-iPSCs did not recapitulate the phenotype of FPD-iPSCs, showing that the mutations were of loss-of-function type. Taken together, this study demonstrated that haploinsufficient RUNX1 allele imposed cell-intrinsic defects on hematopoietic differentiation in human experimental settings and revealed differential impacts of RUNX1 dosage on human and murine megakaryopoiesis. FPD-iPSCs will be a useful tool to investigate mutant RUNX1-mediated molecular processes in hematopoiesis and leukemogenesis.</p>

The Potential of Using Induced Pluripotent Stem Cells in Skin Diseases

<p>The seminal discovery of induced pluripotent stem cells (iPSCs) and subsequent expansion in iPSC research have greatly aided our understanding of stem cell biology and have catapulted the changing face of regenerative medicine into prominence. With easy accessibility and visibility, the skin has been considered a favorable organ to study stem cell behaviors. In particular, the skin provides valuable materials for tissue engineering and testing of novel therapeutic approaches including gene therapy and regenerative medicine. The skin consists of multiple cell lineages, including interfollicular/follicular keratinocytes, melanocytes, sebocytes, and follicular/intradermal mesenchymal cells. Nowadays, iPSCs can be generated from various skin-derived cell populations. Elegant label-tracking experiments have successfully identified their stem/early precursor cell populations. Following molecular and cellular biological experimentations and transgenic studies the elucidation of necessary factors, signaling networks, environments supporting their behavior, and interactions in organogenesis and homeostasis have been uncovered. Taking advantage of the knowledge accumulated by these works, iPSC-derived keratinocytes and melanocytes have been successfully induced, which holds promise as future materials for the bioengineering of the skin and hair, and investigation of biology of skin stem cells. Recently, the concept of cancer stem cells (CSCs) has been proposed. Cancer stem cells have been implied to originate from stem cell subsets and, accordingly, iPSCs may be converted to skin CSCs, especially most life-threatening melanoma CSCs. Those iPSC-derived CSCs may significantly enhance our understanding of cancer biology and allow the development of novel therapeutic strategies. Recently, patient-specific iPSCs were established from intractable genodermatoses, including epidermolysis bullosa, and corrected for their genetic defects. Thus, iPSC technology should open a new era of stem cell biology, genetic/regenerative medicine and oncology in the field of dermatology.</p>
