我が国で頭痛やめまいの症状を持つ患者数は3,000万人を超えると言われており，それに伴う気分の揺らぎに伴うパフォーマンスの低下は，社会全体の活動低下を招いている．これらの患者のなかには，心的ストレスが原因となって身体の不調を訴える症例が多く含まれているが，なぜそのような身体化の症状が生じるかは十分に解明されていない．本研究では，健常者・身体症状症・片頭痛・メニエール病の患者を対象として，自律神経活動，身体状態の変化を感知する内受容感覚，予測的情報処理に基づく信念形成の特性に着目し，気圧変化に対する身体・認知特性を明らかにする．そして，症状改善に向けたプロトコルの開発に取り組む．

脳の広範な機能の理解には神経細胞以外の細胞要素、特に神経細胞と接して存在するグリア細胞の情報を読み出す必要がある。本領域の目標はグリア細胞の状態・機能・細胞間シグナル伝達を包括的に読み出す技術（デコーディング技術）を開発し、脳と身体の間での生体情報の統合を理解する所にある。このような研究を推進するには従来の脳科学研究の成果を踏まえつつも、全く異なるアイディアや計測技術を取り込み、異分野との連携を行う必要がある。総括班ではこのような研究の新規性、技術的優位性、異分野連携に特に力を入れて目標達成に向けた領域の推進に貢献する。

本研究の概要は、神経回路、グリア細胞、血管等の複数の異なる脳内機能要素間を統合するメカニズムを探索する研究である。また、脳内エネルギー代謝は、脳のサバイバルを担うに留まらず、神経回路を流れる情報そのものの特性を左右する存在であることを明らかにする。さらに、本研究では、末梢から中枢へと伸びる迷走神経を、生来の中枢へのアクセスルートとして人為的に活用し、脳内環境を遠隔操作することにも挑戦する。グリア細胞を起点とするエネルギーのスマート供給システムを調整し、神経・精神疾患の治療原理を開拓する。

意欲行動は、行動の開始、行動の持続、目的の達成という段階に分けることができる。意欲行動の持続にかかわる神経基盤は全く分かっていない。予備実験により、腹側線条体へ投射する腹側海馬が、意欲行動の持続に関与するという仮説をたてるに十分な根拠を得た。本研究では、腹側海馬が意欲行動の持続を制御するという仮説を証明するとともに、その神経回路基盤、分子基盤をも明らかにする。

Establishment and applications of model mice with fluctuated deafness by advanced gene-engineering methods

Hearing loss induced by disorder of the cochlea of the inner ear affects ~10% of the global population and thus it provides a great clinical impact. The onset and progress are characterized by different time course―sudden or slow type. Fluctuating type often becomes severe and irreversible deafness. To overcome this deafness type, in the present study we described two deaf animal models by genetic modifications targeting stria vascularis, an epithelial tissue that maintains unique ionic and electrical properties of the cochlea. The mouse line showed slowly progressing hearing loss in response to drug-induced knockdown of an ion transporter in the stria. The other line expressed a light-gated ion channel in the stria and exhibited sudden deafness by optogenetic stimulation. In both mouse types, hearing was recovered after cessation of the perturbation. We also revealed a mechanism underlying hearing loss in each model. This work may contribute to translational research for deafness.

グリアアセンブリによる脳機能発現の制御と病態

事後報告書をまとめるため、研究代表者（池中）と分担者（岡部、尾崎、小泉、田中、竹林）が打ち合わせを行い、その後、メールや電話で相談した。まとめた事後報告書を文部科学省に提出し、平成３０年１０月１日に岡部、小泉、久島（尾崎代理）の３名で事後評価ヒアリングに参加し、岡部がプレゼンテーションを行なった。その結果、Aの評価を得た。その後、研究代表者を池中から竹林へと交代し、業務を滞りなく引き継いだ。事後報告書に基づき、最終報告書の冊子を作成するために本領域の計画班員、公募班員から論文業績、特許、広報などに関する情報を集め、領域の最終評価書のアウトラインを作成した。以上の情報に加え、文科省からの評価書を受領し、５年間の班会議、公開シンポジウム、若手の会などの活動状況、アウトリーチ活動、ホームページを介した広報活動などをまとめ、平成３１年２月下旬に研究代表者と研究分担者で最終報告書を校正・校了した。最終報告書を１６０部印刷し、班員および関係者に広く配布した。なお、代表論文の掲載を含んだ製本を行なうと、ページ数が増えるうえにコストもかかるため、論文業績はタイトル、著者名、雑誌名、巻、ページ番号、doiのみを表記することとした。

Investigation of role of serotonergic neurons in regulation of masseter muscle activity using optogenetics

Various motor behaviors are modulated by serotonin (5-HT). While it is known that both the trigeminal motor nucleus and mesencephalic trigeminal nucleus (MesV) receive serotonergic inputs, it remains unclear how 5-HT affects jaw movement. We examined the effects of 5-HT on masseter (jaw-closing) motoneurons (MMNs) in Wistar rat brainstem slice preparations using a whole-cell recording technique. Activation of 5-HT2A receptors enhanced the function of NMDARs in MMN dendrites and presynaptic 5-HT1B receptors were shown to be involved in inhibition of excitatory synaptic inputs from MesV afferents to MMNs. Our results indicate that optogenetic activation of serotonergic neurons during mastication may promote masticatory movements.

負に立ち向かうウィルパワーをつかさどる神経基盤の解明

ウィルパワーは、負の事象を回避する/すでに起きてしまった負の事象から脱出するという目的を果たすときにも駆動される。本課題では、これまであまり知られてこなかった「嫌なことから逃げるときに駆動されるウィルパワー」の神経基盤を明らかにすることを目的とした。
まず、負に立ち向かうウィルパワーをマウスで定量する評価系 (active avoidance test)の構築に取り組んだ。嫌なこと（電気ショック）を回避するために、どれだけコストを払うのか、オペラント条件付けを利用した行動評価系である。トライアル開始から60秒間ブザーが鳴る。その間に、要求されたレバー回数（5回）を押しきるとブザーが止み、電気ショックを回避できる。5回押すことができなければ、10秒間の電気ショックを受ける。電気ショックが開始されたとしても5回のレバーを押しを達成できれば電気ショックが止まり、ブザー音も止まる。このようにして、電気ショックを回避するためのオペラント行動、電気ショックから脱出するためのオペラント行動を観察する実験系を確立した。
次に、active avoidance testとファイバーフォトメトリー法を組み合わせて、腹側線条体D2-MSNの神経活動変化を記録した。ショックを回避した直後にD2-MSNの活動が低下することが明らかになった。これは負の強化子の獲得直後にD2-MSNの活動が低下すると解釈できる。一方で、通常の餌とレバーを関連づけたオペラント行動においては、餌の獲得、つまり正の強化子の獲得直後にD2-MSNの活動が増加する。いずれも行動を増やす強化子として位置づけられるものの、強化子獲得時の腹側線条体の活動が全く異なることを明らかにした。

Regulatory role of BDNF on emotional and cognitive function

It is well accepted that brain-derived neurotrophic factor (BDNF) is involved in the synaptic plasticity. Human studies demonstrated that serum BDNF amount can be used as a biomarker of neuropsychiatric conditions. However, how BDNF directly regulates brain higher functions is still unknown. Here we developed BDNF gene modifying mice in which BDNF gene expression was capable of time- and region-dependent manipulation. In the stress-free condition, up- or down-regulation of BDNF gene expression did not cause any anxiety-like and depression-like behaviors.In the stressful condition, down-regulation of BDNF gene expression impaired the autonomic function characterized with basal tear secretion. In particular, down-regulation of BDNF expression delayed a recovery from the stress. We conclude that levels of BDNF expression affect the stress resilience in stressful condition but not in stress free condition.
